摘要

目的 结合体内外实验,观察红景天颗粒的抗氧化作用。方法 体内实验中,将昆明种小鼠随机分为5组,即空白对照组、低剂量组、中剂量组、高剂量组及阳性对照组。给药组每天分别灌胃0.1、1、3 g·kg-1的受试物,阳性对照组每天灌胃280 mg·kg-1诺迪康胶囊,空白对照组灌胃等体积去离子水,连续35 d。检测血清、肝、脑组织中SOD、MDA、GSH-PX水平;体外实验,以Vc为对照药,设0.01、0.1、1、10 mg·mL-14个浓度梯度,测定吸光度,计算其清除超氧阴离子自由基和羟自由基的活力。结果 低剂量组可明显升高脑组织超氧化物歧化酶活性水平(P<0.05);中剂量可显著升高血清GSH-PX活力,低剂量组可显著升高脑组织GSH-PX活力(P<0.05);阳性对照药能显著升高肝组织中GSH-PX活力(P<0.05);阳性对照药可显著降低肝组织中MDA水平(P<0.05);体外有清除超氧阴离子和羟自由基作用。结论 红景天颗粒具有体内外抗氧化作用。

Abstract

OBJECTIVE To observe the antioxidative effect of Rhodiola granules in vitro and in vivo. METHODS Female Kunming mice were randomly divided into five groups, ie, blank control group, low-dose group, middle-dose group, high-dose group, and the positive control group. The three treatment groups were given various doses of Rhodiola granules, ie, 0.1, 1, 3 g·kg-1 respectively, the positive control group were given Nuodikang capsules at 280 mg·kg-1, and the blank control group was given deionized water for 35 d. The levels of SOD, MDA, GSH-PX in serum, liver, and brain were determined. For the experiment in vitro, with vitamin C as the control drug, the absorbance of Rhodiola granule solution at concentrations of 0.01, 0.1, 1, 10 mg·mL-1 was measured, and the activity of scavenging superoxide anion free radicals and hydroxyl free radicals was calculated. RESULTS The activity of SOD in the brain of low-dose group mice was significantly increased (P<0.05), the activity of GSH-PX in the serum of middle-dose group mice was significantly increased (P<0.05), the activity of GSH-PX in the brain of low-dose group mice was significantly increased (P<0.05), the activity of GSH-PX in the liver of the positive control group mice was significantly increased (P<0.05), and the level of MDA in the liver of the positive control group mice was significantly decreased (P<0.05). Rhodiola granules could scavenge superoxide anion and hydroxyl free radicals in vitro. CONCLUSION Rhodiola granules have antioxidative effect in vitro and in vivo.
